Alkaline Phosphatase Labeling Kit or Antibody-AP Conjugation Kit enables rapid conjugation of alkaline phosphatase (AP) to an antibody or a protein. The kit includes a modified enzyme (A-AP) and an antibody modifier. The typical coupling efficiency of AP to an antibody is greater than 95%, while the significant coupling is observed within one hour. The kit can also conjugate AP to other proteins.

Alkaline phosphatase labeling kit

1. How to prepare the labeling protein?
The labeling protein must have reactive amine (NH2) groups. It should be purified prior to conjugation. The protein should be prepared in an amine-free buffer. Sodium azide and protein additives should also be avoided. In addition, the desired protein concentration is greater than 2 mg/ml.

2. How much protein can be conjugated?
The kit contains 3 x 100 µgs modified alkaline phosphatase that is sufficient to couple 3x 100 µgs antibody for the optimum result. For coupling of other proteins, the reaction may require to optimize. In addition, the modified enzyme is supplied in the solution form that is convenient for multiple coupling reactions.

3. Is the purification of the conjugate required?
No. Although a small amount of unconjugated enzyme may be present, it can be washed away at the assay step. Therefore, it does not interfere with the assay.
