Click Biosystems Inc. offers a custom biotinylation service (antibody biotinylation). Biotin binds to avidin and streptavidin with high affinity (Kd = 10^–15 M), high specificity, and fast speed. Because of these features, biotin conjugates are ideal reagents for ELISA, western blots, flow cytometry, and surface plasmon resonance (SPR). We have more than 15 years of experience in the generation of biotin conjugates. Our protocol creates high-quality biotin conjugates that are extremely useful for signal amplification in bioassays. Depending on the protein properties, we offer various methods for custom biotinylation of antibodies, proteins, polysaccharides, and oligonucleotides. We also provide custom protein biotinylation on AviTag using ligase BirA. Furthermore, we determine the biotinylation level and molar ratio of biotin to protein.

Antibody Biotinylation Service

Our custom antibody biotinylation service enables rapid biotin labeling of any existing antibodies. The antibody has multiple surface amines that can be labeled. Thus, direct biotinylation may attach multiple sites on the antibody, increasing the detection sensitivity of biotin. In addition, our custom antibody-biotin conjugates contain a hydrophilic spacer to span the antibody and biotin, resulting in great solubility and activity of the product.

Enzymatic Biotinylation of AviTag

Antibodies or proteins engineered with an AviTag (GLNDIFEAQKIEWHE) can attach biotin to the lysine within the peptide tag. Thus, the labeled proteins are homogeneous and can bind to streptavidin for a specified orientation. The biotinylation of AviTag is mediated by the biotin ligase. We will design and add the AviTag to the target protein during the construction. Then, the biotinylation is carried out in vitro or co-expressed in E. coli with the target protein. The customer can also submit an existing protein engineered with the AviTag for in vitro biotin labeling. Additionally, our products are free of enzyme contamination.
