The synthetic BRAF 600V DNA is a linear plasmid that contains a 773 bp DNA fragment of the human BRAF wild-type gene. The DNA insert covers the BRAF-600V (GTG) wild-type codon 600 and 5′- and 3′-flanking regions. The BRAF 600V (GTG) wild-type plasmid is verified by Sanger sequencing and linearized.

Mutant BRAF has been implicated in the pathogenesis of several cancers, including melanoma, non-small cell lung cancer, colorectal cancer, papillary thyroid cancer, and ovarian cancer. BRAF is altered in 6.01% of all cancers with colon adenocarcinoma, cutaneous melanoma, lung adenocarcinoma, melanoma, and thyroid gland papillary carcinoma having the greatest prevalence of alterations. In addition, BRAF is a predictive biomarker for use of drugs and antibodies in patients. Melanoma, colorectal carcinoma, non-small cell lung carcinoma, ganglioglioma, and gastrointestinal stromal tumor have the most therapies with BRAF as a predictive biomarker.
