The synthetic BRAF V600K DNA is a linear plasmid that contains a 773 bp DNA fragment of the human BRAF-V600K (AAG) gene. The DNA insert covers the BRAF-V600K (AAG) mutant codon and 5′- and 3′-flanking regions. The BRAF V600K (AAG) plasmid is verified by Sanger sequencing and linearized.

BRAF V600K is present in 0.16% of AACR GENIE cases, with melanoma, cutaneous melanoma, melanoma of unknown primary, esophageal melanoma, and lung adenocarcinoma having the greatest prevalence. Thus, BRAF V600K is a predictive biomarker for use of dabrafenib, vemurafenib, trametinib, binimetinib, cobimetinib, and encorafenib in patients.
