The synthetic BRAF V600R DNA is a linear plasmid that contains a 773 bp DNA fragment of the human BRAF-V600R (AGG) gene. The DNA insert covers the BRAF-V600R (AGG) mutant codon and 5′- and 3′-flanking regions. The BRAF V600R (AGG) plasmid is verified by Sanger sequencing and linearized.

BRAF V600R is present in 0.04% of AACR GENIE cases, with melanoma, cutaneous melanoma, melanoma of unknown primary, pancreatic adenocarcinoma, and duodenal adenocarcinoma having the greatest prevalence. In addition, BRAF V600R is a predictive biomarker for use of dabrafenib, vemurafenib, binimetinib, cobimetinib, encorafenib, and trametinib in patients. Of the therapies, melanoma has the most therapies targeted against BRAF V600R or its related pathways.
