Streptavidin-oligo conjugates are useful molecular connectors in the DNA-directed immobilization (DDI) of biotinylated antibodies using DNA microarrays as immobilization matrices. A new or interesting marker can be identified by a streptavidin-oligo conjugate without the need for custom antibodies or performing the coupling yourself.

The oligonucleotide is normally conjugated to amines (lysine residues and n-terminus) on streptavidin. Such a random coupling method may impair the function of streptavidin. In addition, streptavidin contains five amino groups and can conjugate more than one oligo. Thus, the product is normally heterogeneous. In contrast, we use a site-specific conjugation method to generate our streptavidin-oligo conjugates. Our engineered streptavidin-azide has a unique azido group at the c-terminus that allows site-specific attachment of the oligonucleotide. In addition, we use a long hydrophilic spacer to span streptavidin and oligo. Moreover, we couple one oligo to each streptavidin molecule. Thus, our streptavidin-oligo conjugates are homogeneous and retain the full function of streptavidin.
