Product Overview

HRV3C Protease (GST-tag, His-tag) is a recombinant form of human rhinovirus (HRV) type 14 3C protease. The enzyme recognizes a specific sequence LEVLFQ/GP and cleaves between Gln (Q) and Gly (G). Our HRV3C Protease (GST-tag, His-tag) is useful for removal of tag sequences from proteins having the cleavage site. It can also cleave small peptides with the processing site. Moreover, our HRV3C protease has both His-tag and GST-tag, allowing easy removal from the sample after cleavage.

HRV3C Protease (GST-tag, His-tag) has great stability and robust activity at 4°C. In addition, it is active in buffers that are suitable for most proteins. Typically, 1μg of the HRV3C Protease (GST-tag, His-tag) can cleave 100 μg of target protein at 4°C. However, the cleavage efficiency may vary due to the sequences around the cleavage site and conformation of the target protein. More enzyme, longer cleavage time, or higher temperature (37°C) may enhance cleavage efficiency.

Cleavage of Protein
1. Prepare the target protein (1-2 mg/ml) in cleavage buffer by dialysis. An example of cleavage buffer is 50 mM Tris-HCl, pH 8.0, 150 NaCl, 1 mM DTT.
2. Add HRV3C Protease to the target protein. 1000 units of HRV3C Protease can cleave 100 mg of target protein. Other Protease-to-target protein ratios (w/w) of 1:50 to 1:200 should work for most target proteins.
3. Incubate the cleavage reaction overnight at 4°C for complete cleavage.
