Description
The synthetic BRAF DNA (Codon 600) products are cell-free linear plasmids that contain a 773 bp DNA fragment of the human BRAF gene. The DNA insert covers the BRAF codon 600 wild-type or mutations. It also has 5′ and 3′-flanking regions. The BRAF plasmid DNA products are verified by Sanger sequencing and linearized.
The BRAF gene encodes the protein serine/threonine-protein kinase B-raf. It plays a critical role in cell proliferation, differentiation, and transcriptional regulation. The BRAF gene is altered in more than 6% of all cancers with colon adenocarcinoma, cutaneous melanoma, lung adenocarcinoma, melanoma, and thyroid gland papillary carcinoma.
BRAF is also a predictive biomarker for use of many drugs, such as vemurafenib and panitumumab in patients. BRAF V600E, BRAF Mutation, BRAF Codon 600 Missense, BRAF V600K, and BRAF K601E are the top alterations on BRAF targeted by therapies. Melanoma, colorectal carcinoma, non-small cell lung carcinoma, ganglioglioma, and gastrointestinal stromal tumor have the most therapies with BRAF as a predictive biomarker. The synthetic BRAF DNA products can be used as the standard reference of the wild-type codon 600 and its mutations.
